Figure D. Synthesis of multiple cytokine and inducible genes is strongly reduced or absent in SCID T cells.

T cell lines from patients (P1, P2) and controls (Co1, Co2) were left unstimulated (lanes 1-4) or stimulated for 4h with PMA plus ionomycin (lanes 5-8) and cytokine levels were assessed by RNase protection assay. The respective amounts of CD4+ / CD8+ cells in the T cell lines used were as follows: Co1 79/5%, Co2 5/80%, P1: 5/89%, P2 68/25%. GAPDH and L32 are housekeeping genes as control for equal loading of lanes. IL, interleukin; IFN, interferon; TNF, tumor necrosis factor; (G)M-CSF, (granulocyte) macrophage colony stimulating factor; MIP, macrophage inflammatory protein; FasL; Fas ligand; Ltn, lymphotactin.
